Carlo first started his showbiz career when he became a part of the comedy variety show Ang TV back in 1992. 

Carlo rose into fame for his portrayal of Ojie in the 1998 film Bata, Bata…Pa'no Ka Ginawa? alongside veteran actress Vilma Santos. 

Projects were pouring in for Carlo who was then given a role in the youth-oriented program called G-mik where he was paired with Angelica Panganiban from 1999 to 2002.

While doing G-mik, he also went on to star in another Star Cinema classic film called Daddy O! Baby O! with King of Comedy Dolphy during the year 2000. 

In 2003, he played the role of Tri Montecillo in the comedy film Ang Tanging Ina, which later on had a sequel called Ang Tanging Ina Ninyong Lahat in 2008 and Ang Tanging Ina Mo Rin (Last na 'To!) in 2010. 

Carlo was also given the role of Oman in the television series Bituing Walang Ningning together with Sarah Geronimo.

He continued to do Kapamilya projects like the teleserye Annaliza, which aired from 2013 to 2014. 

Carlo once again proved his talent in acting in the heartwarming MMK episode titled "Wedding Gown." 

Soon after, the actor landed the role of Arnold Magsakay in the TV series FlordeLiza starring Jolina Magdangal and Marvin Agustin. 

He was also seen in several Ipaglaban Mo episodes.

In 2018, Carlo Aquino and Angelica Panganiban made their long-awaited reunion movie called Exes Baggage, which is about a former couple who are re-acquainted and try to heal the wounds from the past. 

The Kapamilya actor also headlined the movies Meet Me in St. Gallen with Bela Padilla, as well as the movie Ulan with Nadine Lustre. 

He also played the role of Gali Pastrano in Isa Pa with Feelings, a deaf-mute and a sign-language teacher to Mara, who was played by Maine Mendoza. 

From 2018 to 2019, Carlo got a role in the TV series Playhouse starring Angelica Panganiban and Zanjoe Marudo. 

During the pandemic, Carlo was still lucky to star in the TV series A Soldier's Heart as Sgt. Abraham Kamlum. 

In 2021, the actor was given the role of Jordan together with Erich Gonzales in the series La Vida Lena, which is still airing. 

Recently, Carlo shared a missed opportunity when he was offered a role in the hit Korean series Squid Game.

Carlo later on explained that it was the travel restrictions because of the pandemic that hindered him from going to Korea and do his scenes for the hit series.
